Postdoctoral Position In Cryo-Electron Tomography Of Chromatin Organization In Situ

An exciting opportunity exists for a postdoctoral researcher to work on in situ chromatin organization using in situ cryo-electron tomography, fully-funded for at least two years.
Chromatin, which plays a crucial role in controlling gene expression and maintaining the genome, is one of the most enigmatic and extraordinary substances of living matter. For instance, it is well soluble but is subject to phase separation; it behaves like a solid and a fluid at different scales; it can be dense but transparent to diffusion; it combines hierarchical organization with an extreme structural variability. Despite decades of research, our understanding of chromatin structure beyond the nucleosome level is fragmented, due to the insufficient resolution of in situ imaging and the limited applicability of conventional averaging tools. However, recent developments in cryo-electron tomography and continuous conformational variability analysis have the potential to overcome these limitations and decipher the structure of chromatin in situ, in its functional environment.
If successful, you will be part of the Cryochrom collaborative project (https://anr.fr/Project-ANR-20-CE11-0020), which brings together research groups with long-standing experience and expertise in cryo-electron microscopy and chromatin tomography, image processing and in silico modelling.
As part of the CBI (http://www.igbmc.fr/grandesstructures/cbi/), located in vibrant and dynamic Strasbourg, you will work in a state-of-the-art scientific and technological environment in integrated structural biology. The CBI hosts the French and European Infrastructures for Integrated Structural Biology, FRISBI (http://frisbi.eu/); Instruct-ERIC (https://www.structuralbiology.eu/); and iNext-Discovery (https://inext-discovery.eu/network/inext-d/home). It is equipped with cutting-edge cryo electron microscopes ( Titan Krios and Glacios), and a cryo focused ion beam scanning electron microscope (cryo-FIB/SEM). There is also CLEM module (Leica Microsystems) available for correlative cryo-fluorescent imaging and integrated sample transfer to FIB/SEM or cryo-TEM. In addition, the cryo-EM platform has a suite of associated equipment for sample preparation and dedicated computing resources for image processing and 3D reconstruction.
You should have a PhD degree in structural biology, biophysics or a related field. Experience in cryo-electron tomography and/or tomographic image processing is essential. Hand-on skills with cryo-FIB-SEM or CEMOVIS are an advantage.
