We are interested in the cellular and molecular mechanisms controlling the differentiation and the function of the immune cells. We use a simple genetic model to perform in vivo and in vitro analyses that would be otherwise not possible/unaffordable in more complex animals.
Our team has dissected the transcriptional pathways expressed in Drosophila glia and macrophages, the immune cells present within and outside the nervous system. Beyond their role in the defense against pathogens and inflammation, they both connect different cells and macrophages connect even distant tissues, working as sensors and signaling hubs of the internal state in physiological conditions. They have also been recently shown to be major actors in disease, from cancer to neurodegeneration, and therefore represent powerful therapeutic targets. The recent advances in high throughput technologies allow us to study these cells at unprecedented resolution and to integrate the inter-tissue interactions that allow homeostasis. This opens challenging perspectives to understand the mechanisms controlling immune cell development and physiology.
Using a multidisciplinary approach going from genetics and bioinformatics to molecular and cellular biology, we aim at understanding:
- Macrophage origin and heterogeneity
- The impact of macrophages in development, physiology and pathology, especially in the context of neuro-immunity
- The role and mode of action of an evolutionarily conserved anti-inflammatory transcriptional cascade
We are a very international team, with young investigators from France and abroad as well as many collaborators visiting the laboratory to learn techniques and perform experiments. We benefit from the facilities present at the IGBMC to achieve our goals.
Activités
Conduite et analyse d'expériences de biologie moléculaire (PCR, FISH, clonage, western blot) et cellulaire, microscopie confocale incluse
Dissection et préparation d'échantillons (pour immunomarquage, in situ, etc)
Soutient technique pour la construction de souches de drosophile (amplification de souche, croisements, sélection de génotypes)
Gestion des commandes et stocks du laboratoire
Aide au maintient des stocks de drosophile
Accompagnement de l’activité des stagiaires
Compétences techniques
Connaissance générale en biologie moléculaire (PCR, FISH, clonage, immunomarquages, western blot)
Connaissance générale en culture cellulaire
Connaissance de base en génétique et en anatomie recommandées mais pas essentielles
Langue anglaise compréhension écrite et orale (A2 à B1)
Compétences informatiques
Maîtrise de l'outil bureautique et informatique (Pack office)
Savoir communiquer et rendre compte
Savoir travailler en équipe
Savoir s’adapter et être polyvalent(e)
Suivre les bonnes pratiques de laboratoire
Tenir rigoureusement un cahier de laboratoire (Labguru)
