Protein‐protein interactions are essential for the proper functioning of most cellular processes and their deregulation can cause disease. The last years have seen major achievements in the systematic mapping of human protein interactions, yet information on how these proteins interact with each other is lacking hindering understanding of the molecular function of these interactions and which mutations would perturb these interactions and cause disease. I will present on our efforts to develop various tools for the prediction of protein interaction interfaces including recent results on the benchmarking of AlphaFold Multimer and the application of these tools to interactions involving disease‐associated proteins followed by experimental validation.
