Phage and Yeast united for the better: Synthetic Nanobody® selection and optimization

Phage display nanobody selection from our fully synthetic humanized library is a reality with more than hundred successful projects since the publication of our library developed with Sandrine Moutel, Aurélien Olichon and Franck Perez1.

We selected with our colleagues and customers VHHs against unstructured proteins 2, peptide tag3, various soluble proteins4, small molecules1 or challenging DNA structures5.

Building from our 25 years of experience in Yeast genetic, we developed a yeast-two-hybrid and a Yeast display platform for intrabodization, and optimization of nanobodies coming from our library or from the lama6.

Working fluently between phage and yeast world, we can address many complex projects particularly when there is bottleneck in antigen expression or purification. We will illustrate that screening of naïve yeast library is also a very powerful alternative to find intracellularly functional VHHs against soluble antigens.

We will present new results around nanobodies active against TAU aggregation, nanobody against V5 tag and Ubiquitin and chain.
