Congratulations to Quentin Giraud for the acceptance of his thesis ! His work was directed by Jocelyn Laporte and titled: "Characterization of new therapeutic approaches for centronuclear myopathies".

credits: Théo Brisset, IGBMC

During his thesis, Quentin Giraud worked on centronuclear myopathy to help develop gene therapies and understand the underlying mechanisms. In a mouse model of BIN1-related centronuclear myopathy, he overexpressed MTM1 phosphatase in skeletal muscle. With a neonatal injection, he showed an improvement in muscle strength and cellular organization. A late injection showed a therapeutic effect, but to a lesser extent. He also determined the therapeutic mode of action of MTM1, relying on phosphatase-independent functions. Subsequently, he also characterized the most suitable BIN1 isoform for a gene replacement therapy approach, as well as potential limitations of this treatment.

During his time at the IGBMC, the PhD student has strengthened his understanding of the gene therapy development process and of working with the animal model.

Thanks to our different platforms for their support in this work ! Thanks to ICS’ different services and to our molecular biology platform.

Thanks to AFM-Téléthon and FRM for their support. Quentin Giraud’s work was partially financed through the program “Espoir de la recherche” of the Fondation pour la Recherche Médicale.

Quentin Giraud will soon be joining the Karolinska Institutet in Sweden, as a post-doctorant, where he will be investigating the genetic mechanisms responsible for ageing ! We wish him good fortune in the next part of his career !
