Structure-function relationships of nuclear receptors and their co-regulators

Nuclear receptors are ligand-regulated transcription factors that control nearly all aspects of human physiology. We are studying several members of the nuclear receptor family including the vitamin D receptor (VDR), the peroxisome-activated receptors (PPARs) and the retinoid receptors (RARs and RXRs). A major interest of the group is to understand how the activation and repression of nuclear receptor transcription are regulated at the structural and molecular level using experimental methods combining structural, biophysical and cellular approaches. Our work concerns ligand recognition, allosteric control of nuclear receptor activity and coregulators recognition in a physiological and pathological context. In addition, we characterize important coregulators involved in the control of transcription (Mediator, CBP/p300) and their dysfunction in a pathological context. These studies provide important information to propose new therapeutic strategies and to design new therapeutic agents with selective activities.
